mRNA递送载体(脂质纳米粒)合成砌块
2023 年10月2日诺贝尔生理学或医学奖授予美国科学家卡塔林·卡里科(Katalin Karikó)和德鲁·魏斯曼(Drew Weissman),以表彰他们在核苷碱基修饰方面的发现,开发了针对新冠感染的mRNA(简称核酸)疫苗。mRNA是一个带负电的长链大分子,细胞膜也带有负电,静电排斥作用使mRNA难以进入细胞内,而脂质纳米粒(lipid nanoparticles, LNPs)作为多种治疗剂的常用载体,在有效保护mRNA并将其运输到细胞当中发挥了至关重要的作用。
LNPs是一种多组分系统,通常由阳离子脂质(可电离脂质)、辅助脂质、胆固醇、保护剂聚乙二醇-脂质共轭物组成(如下图所示),其中阳离子脂质与带负电荷的mRNA结合,将其转变为中性,不仅限制LNPs的毒性,同时促进LNPs中的核酸包封;辅助脂质、胆固醇和聚乙二醇-脂质可以维持粒子的结构并提高稳定性。
国外有“mRNA 疫苗研发三巨头”:Moderna、CureVac、BioNTech,均采用脂质纳米技术递送,这也是目前在核酸药物领域应用比较广泛的递送技术类型。Moderna的SM-102和BioNTech的Acuitas ALC-0315具有一些共同的特征,包括叔胺,支链尾和酯接头,下表根据现有的专利和文献显示了可能的脂质结构。
